 Pengertian pewarnaan
● Pengenalan bentuk mikroba, kecuali mikroalgae
harus dilakukan perwanaan terlebih dahulu agar
dapat diamati dengan jelas.Tujuan dari
pewarnaan adalah untuk mempermudah
pengamatan bentuk sel bakteri, dan melihat
reaksi jasad terhadap pewarnaan yang di berikan
sehingga sifat fisik atau kimia jasad dapat
diketahui (Hadioetomo,1991).

Pewarnaan bakteri pada umumnya bertujuan untuk
mempermudah dalam pengamatan morfologi
bakteri dengan bantuan mikroskop.
Bakteri umumnya tidak berwarna dan hampir tidak
terlihat karena kurang kontras dengan air dimana
mereka berada.
Pewarnaan sangat dibutuhkan untuk melihat
morfologi bakteri baik untuk pengamatan
intraseluler maupun morfologi keseluruhan.

Secara Garis Besar
1. Pewarnaan bakteri hidup
- menggunakan bahan warna yang tidak toksis
- dilakukan untuk melihat pergerakan bakteri
- pemeriksaannya dilakukan dengan menggunakan
pipet tetes gantung
2. Pewarnaan bakteri mati (fixed state)
- bertujuan untuk melihat struktur luar atau dalam
bakteri
-memperjelas ukuran bakteri dan
-melihat reaksi bakteri terhadap warna yang diberikan,
sehingga dapat diketahui sifat fisik dan kimia dari
bakteri tersebut.

Contoh pewarnanya : Methylene blue, Basic fuchsin
dan Crystal violet
Pewarna bekerja dengan baik pada bakteri karena
bersifat basa dan alkalin. Pewarnaan sederhana
bertujuan untuk memberikan kontras antara bakteri
dan latar belakang

 2. Pewarnaan Negatif
Pewarnaan yang menggunakan pewarna asam
Contoh pewarnanya : Eosin, Negrosin dan Tinta cina sebagai pewarna utama
Dilakukan pada bakteri yang sukar diwarnai oleh pewarna sederhana.
Pewarnaan negatif bertujuan untuk memberi warna gelap pada latar
belakang dan tidak memberi warna pada sel bakteri
Pewarna yang digunakan adalah pewarna asam dan memiliki komponen
kromoforik yang bermuatan negatif, yang juga dimiliki oleh sitoplasma
bakteri.
Pada pewarnaan negatif ini sel bakteri terlihat transparan (tembus pandang)
