Satu siklus sel terdiri dari beberapa fase yang sangat
terkoordinasi antara satu fase dengan fase yang
lain :
Pertumbuhan sel
Replikasi DNA
Duplikasi kromosom dan distribusinya ke sel baru
Pembelahan sel/sitokinesis/cleavage
PROKARIOTA :
Pertumbuhan dan replikasi DNA terjadi sepanjang siklus sel
(tidak ada stadium siklus sel), duplikasi kromosom dan
distribusinya ke sel generasi berikutnya dilakukan melalui
asosiasi kromosom dengan membran sel pada waktu
segmentasi sel.
EUKARIOTA:
Tiap fase merupakan satu periode siklus sel. Pengaturan
siklus sel dipengaruhi oleh growth factor sehingga
pembelahan dan proliferasi sel disesuaikan dengan
kebutuhan fisiologi (homeostatis)

INTERFASE terdiri dari :
Fase G-1 (GAP-1) :
Interval antara akhir M dan permulaan S- persiapan memasuki fase
S, sel dalam keadaan aktif melakukan biosintesis untuk
pertumbuhan sel selain replikasi DNA. Pada akhir fase S masa sel
menjadi 2x sel awal. Fase G1 bervariasi pada setiap jenis sel.
Fase S (SINTESIS) :
Sel mensintesis DNA dengan replikasi DNA nukleus menjadi 2 x
mula-mula pada akhir fase S.
Fase G2 (GAP 2) :
Interval antara akhir fase S dan awal mitosis (fase M) persiapan
memasuki fase M, sel mempersiapkan perangkat untuk mitosis
seperti biosintesis histon dan tubulin.
TELOFASE
Kromatid tunggal sampai di kutub masing2
Membran inti terbentuk kembali di sekitar kromatin, nukleus,
nukleolus mulai tampak
Kromatid terurai kembali menjadi panjang & tipis membentuk
kromatin halus
Sitokinesis berlangsung sehingga dihasilkan 2 sel anak yang
identik (Furrowing)
SITOKINESIS :
Sitokinesis melalui proses yang disebut cleavage yang
dibentuk cincin kontraktil yang membagi sel menjadi 2
bagian.
Cincin kontraktil terdiri dari aktin dan miosin yang
berasosiasi dengan membran plasma yang berbatasan
dengan sitoplasma.
Cleavage biasanya akan menghasilkan 2 sel baru yang
identik dan simetri.
Pengatur (Pengontrol) Siklus Sel :
1. Maturation Promoting Factor (MPF)
2. Start Point : START (Saat masuk fase S)
Maturation Promoting Factor (MPF)
suatu protein kompleks yang terdiri dari 2 sub unit :
1. cdc2
Cdc terdapat dalam bentuk inaktif pada interfase, dapat
diaktifkan oleh cyclin selama mitosis & fosforilasi serine
dan threonin
2. Cyclin
protein yang disintesis selama interfase dan didegradasi
selama mitosis (M & G-cyclin).
Cyclin bergabung dengan Cdc-P (inaktif)
Kompleks Cdc-cyclin menfosforilasi protein target,
sehingga terjadi :
Kondensasi kromosom
Fragmentasi membran nukleus/lamin
Perubahan sel hingga siap mitosis
Assembly membran inti pada waktu interfase.

Pada interfase :
.Sentriol dalam sentrosom membelah.
.Organel dalam sitoplasma yang memulai pembelahan
sentrosom & mengatur komponen (aparatus) mitotik (aster,
spindel, traction fiber)

a. Diploid (2n=4 kromosom)

5. Mitosis tak terkendali : Kanker

Tujuan Meiosis :
Mendapatkan individu yang memiliki jumlah kromosom
normal (46) yang berasal dari ayah dan ibu

The result is two genetically identical sister chromatids which
remain attached at their centromeres
Prophase I R
This is a crucial phase for miosis.
During this phase each pair of
chromatids dont move to the equator
alone, they match up with their
homologous pair and fasten together
(synapsis) in a group of four called a
tetrad.
Extremely IMPORTANT!!! It is during
this phase that crossing over can
occur.
Crossing Over is the exchange of
segments during synapsis.
Leptoten/Leptonema (tahap benang-benang tipis):
kromosom yang panjang dan tipis mulai berkondensasi
Zigoten/zigonema:
pasangan kromosom homolog bertemu dan digabungkan oleh
sebuah struktur protein seperti pita yang disebut kompleks
sinaptonema (awal sinapsis)
Pakhiten/pakhinema (tahap benang tebal):
sinapsis sudah terbentuk dan nodul-nodul rekombinasi mulai
muncul di sepanjang kromosom-kromosom yang bersinapsis.
Terjadi proses crossing over (pindah silang) sehingga
menghasilkan pertukaran materi genetik. Titik pertukaran disebut
kiasmata (tunggal : kiasma). Hanya 2 dari 4 kromatid yang
crossing over.
Diploten/diplonema (tahap benang-ganda):
Tahap ini dimulai ketika kompleks sinaptonema mulai menghilang
sehingga kromatid-kromatid dan kiasmata mudah terlihat.
Diakinesis (tahap pergerakan ganda) :
Kromosom mencapai kondensasi maksimal, sedangkan nucleolus
dan membrane nucleus menghilang.
