PENDAHULUAN
A. Latar Belakang
Bila radiasi infra merah dilewatkan melalui suatu cuplikan, maka molekul-
molekulnya dapat menyerap (mengabsorpsi) energi dan terjadilah transisi diantara tingkat
vibrasi (ground state) dan tingkat vibrasi tereksitasi (excited state). Contoh suatu ikatan
C – H yang bervibrasi 90 triliun kali dalam satu detik harus menyerap radiasi infra merah
pada frekuensi tersebut (9,0 x 10 13 Hz, 3000 cm –1) untuk pindah ke tingkat vibrasi
tereksitasi pertama. Pengabsorpsian energi pada berbagai frekuensi dapat dideteksi oleh
spektrofotometer infrared, yang memplot jumlah radiasi infra merah yang diteruskan
melalui cuplikan sebagai fungsi frekuensi (atau panjang gelombang) radiasi. Plot tersebut
adalah spektrum infra merah yang memberikan informasi penting tentang gugus
fungsional suatu molekul.
Spektrofotometer FTIR merupakan alat yang dapat digunakan untuk identifikasi senyawa,
khususnya senyawa organik, baik secara kualitatif maupun kuantitatif.

a. Analisis kualitatif
Analisis kualitatif dengan spektroskopi FTIR secara umum digunakan untuk identifikasi
gugus-gugus fungsional yang terdapat dalam suatu senyawa yang dianalisis (Silverstein dan
Bassler, 1998).
b. Analisis kuantitatif
Analisis kuantitatif dengan spektroskopi FTIR secara umum digunakan untuk menentukan
konsentrasi analit dalam sampel. Analisis kuantitatif dengan FTIR digunakan hukum Lambert
Beer’s. Hukum Lambert Beer’s dinyatakan sebagai berikut:

PEMBAHASAN
Fourier Transformed Infrared (FTIR) merupakan salah satu alat atau instrument
yang dapat digunakan untuk mendeteksi gugus fungsi, mengidentifikasi senyawa dan
menganalisis campuran dari sampel yang dianalisis tanpa merusak sampel. Daerah
inframerah pada spektrum gelombang elektromagnetik dimulai dari panjang

gelombang 14000 cm-1 hingga 10-1. Berdasarkan panjang gelombang tersebut
daerah inframerah dibagi menjadi tiga daerah, yaitu IR dekat (14000-4000 cm -1) yang
peka terhadap vibrasi overtone, IR sedang (4000-400 cm-1) berkaitan dengan transisi
energi vibrasi dari molekul yang memberikan informasi mengenai gugus-gugus
fungsi dalam molekul tersebut, dan IR jauh (400-10 cm-1) untuk menganalisis
molekul yang mengandung atom-atom berat seperti senyawa anorganik tapi butuh
teknik khusus (Schechter, 1997; Griffiths dan Chalmers, 1999). Biasanya analisis
senyawa dilakukan pada daerah IR sedang (Tanaka dkk, 2008).

ikatan pada gugus fungsi adalah:

Berdasarkan pembagian daerah panjang gelombang sinar inframerah dibagi atas
tiga daerah yaitu pada Tabel 1.

1. The source: energi IR yang dipancarkan dari sebuah benda hitam menyala.
Balok ini melewati melalui logam yang mengontrol jumlah energi yang
diberikan kepada sampel.
2. Interoferometer: sinar memasuk iinterferometer, spectraencoding ‟mengambil
tempat, kemudian sinyal yang dihasilkan keluar dari interferogram.
3. Sample : sinar memasuki kompartemen sampel dimana diteruskan melalui
cermin dari permukaan sampel yang tergantung pada jenis analisis.
4. Detector: sinar akhirnya lolos ke detektor untuk pengukuran akhir. Detektor ini
digunakan khusus dirancang untuk mengukur sinar interfrogram khusus.
Detektor yang digunakan dalam Spektrofotometer Fourier Transform Infra Red
adalah Tetra Glycerine Sulphate (disingkat TGS) atau Mercury Cadmium
Telluride (disingkat MCT). Detektor MCT lebih banyak digunakan karena
memiliki beberapa kelebihan dibandingkan detektor TGS, yaitu memberikan
respon yang lebih baik pada frekwensi modulasi tinggi, lebih sensitif, lebih
cepat, tidak dipengaruhi oleh temperatur, sangat selektif terhadap energy vibrasi
yang diterima dari radiasi infra merah.
5. Computer: sinyal diukur secara digital dan dikirim ke komputer untuk diolah
oleh Fourier Transformation berada. Spektrum disajikan untuk interpretasi
lebih lanjut.
Teknik Transmisi
Teknik transmisi dalam memperoleh spektra IR, yaitu dengan melewatkan berkas
sinar inframerah melalui sampel. Kelebihan dari teknik ini yaitu spektra transmisi
mempunyai rasio sinyal noise yang tinggi, relatif tidak mahal, dan bersifat universal
karena bekerja pada sampel padat, cair, gas dan polimer. Kekurangannya yaitu pada

sampel yang dianalisis, idealnya harus berketebalan 1-20 mikron dan penyiapan
sampel membutuhkan waktu yang lama. Teknik transmisi dibagi menjadi tiga yaitu:
(a) spektra transmisi sampel padat, (b) spektra transmisi sampel cairan, dan (c)
spektra transmisi sampel gas.

Teknik Reflektan (Pantulan)
Teknik reflektan digunakan untuk sampel yang sulit untuk dianalisis dengan
teknik transmitan. Teknik pantulan atau reflectance ini sinar inframerah kembali
dipantulkan paa sampel yang dianalisis. Keuntungan dari teknik reflektan adalah
preparasi sampel cepat, mudah, tidak dipengaruhi oleh ketebalan sampel dan non-
destrusi. Terdapat 2 metode reflektan, yaitu:
Teknik ATR digunakan untuk memperoleh spektra zat padat, cair, semipadat,
dan lapisan tipis. ATR dilakukan dengan menggunakan aksesori dalam kompartemen
sampel spektrofotometer FTIR. Bagian inti aksesori ATR terlihat kristal (berupa
bahan transparan inframerah) dengan indeks bias yang tinggi (Rohman, 2014;
Beasley, et al., 2014). ATR merupakan teknik yang baik dan tangguh untuk sampling
IR. ATR berguna untuk sampling permukaan bahan yang halus, dimana bahan
tersebut sangat tebal atau buram untuk pengukuran IR dengan transmisi. Teknik ATR
bersifat nondestruktif, preparasi sampel sedikit dan tidak memerlukan preparasi
sampel, dan cepat. ATR dapat mengukur padatan (seperti kertas, kaca, keju, daging,
serbuk dan bahan yang warna gelap) dan cairan termasuk larutan non-aqueous
(seperti minyak dan pasta), polimer dan bahan organik lain (Sun, 2008).
Teknik ini merupakan salah satu jenis reflektan eksternal. Dalam pantulan
eksternal, radiasi yang mengenai difokuskan ke sampel dan 2 bentuk pantulan dapat
terjadi, yakni: (1) specular atau pemantulan yang terjadi pada permukaan yang
mengilap; dan (2) diffuse (menyebar). Reflektan eksternal mengukur radiasi yang
dipantulkan dari sebuah permukaan. Bahan yang digunakan harus memantul
atau diikatkan dengan bahan yang memantul.

1. Pantulan specular terjadi ketika sudut radiasi yang dipantulkan sama dengan
sudut datang. Banyaknya sinar yang dipantulkan tergantung pada sudut
datang, indeks bias, kekasaran permukaan, dan sifat-sifat serapan sampel.
Pada teknik ini, sampel-sampel padat diletakkan pada permukaan yang rata.
Teknik specular umum digunakan untuk sampel-sampel cairan yang dapat
dituang melalui teflon.
2. Pada spektroskopi pantulan menyebar, energi yang menetrasikan satu atau
lebih partikel dipantulkan ke segala arah dan komponen ini disebut dengan
pantulan menyebar (diffuse reflectance). Dalam teknik inframerah, pantulan
menyebar (biasa dikenal dengan DRIFT = Diffuse Reflectance Infrared
Fourier Transform). Pada teknik pantulan menyebar, serbuk sampel dicampur
dengan serbuk KBr, tetapi tidak memerlukan pembuatan pelet dengan tekanan
hidrofilik. DRIFT sangat sesuai untuk sampel-sampel serbuk dan serat.

A. Kesimpulan
1. Spektroskopi inframerah merupakan suatu metode yang mengamati interaksi
molekul dengan radiasi elektromagnetik yang berada pada daerah panjang
gelombang 0.75-1.000 µm.
2. Spektroskopi inframerah sebuah metode analisis instrumentasi pada senyawa
kimia yang menggunakan radiasi sinar infra merah. Spektroskopi inframerah
dugunakan untukmengidentifikasi gugus fungsi yang terdapat dalam
senyawa organik.

The objective of this research is to investigate number and types of bioactive
compounds present in Aloe Vera buds grown in North East Punjab region of
Pakistan by FTIR which is simple, nondestructive, cost effective and user friendly.
Aloe vera was cultivated under normal atmospheric conditions. After two years,
buds (early stage of flowers) were grown on twigs. The buds were plucked and
blended with distilled water in National Juicer machine to have a concentrated
blend. The blend was filtered to get clear solution, mixed with ethyl acetate and
then solvent extracted. The organic part was isolated and dried at water bath at
60°C. Dried sample was analyzed using FTIR spectroscopic analysis. All

4. CONCLUSION

of both samples especially in the higher
frequency region and assigned to

matrix.
