STANDAR OPERASIONAL PROSEDUR (SOP)

SPEKTROFOTOMETER GENESIS 10S UV-Vis

1. Pastikan alat spektrofotometer tersambung dengan sumber listrik

2. Tekan tombol on/off yang terletak pada bagian belakang spektrofotometer
3. Masukkan kuvet yang berisi sampel pada cell holder dan kuvet blanko pada cell holder
dengan tanda B
4. Tutup cell holder
5. Tekan tombol B pada papan tombol
6. Tekan tombol dibawah tulisan measure blank
7. Tunggu sampai di layar monitor muncul 0.000 A
8. Kemudian tekan tombol test sehingga akan muncul beberapa menu
9. Atur panjang gelombang cahaya yang diinginkan pada menu wavelenght sesuai dengan
keinginan
10. Atur jumlah sampel yang diuji pada menu number of sample
11. Kemudian tekan tombol test
12. Lalu tekan tombol run test
13. Tunggu hingga hasil muncul
14. Catat hasil yang didapatkan kemudian tekan tombol esc hingga angka 0,000 A muncul
di layar monitor
15. Buka tutup cell holder kemudian keluarkan kuvet yang berisi sampel dan blanko
16. Tutup kembali cell holder
17. Tekan tombol on/off di belakang spektrofotometer
18. Lepaskan kabel spektrofotometer yang tersambung dengan sumber listrik
Hal hal yang perlu diperhatikan dalam pemakaian spektrofotometer
1. Pastikan kuvet yang digunakan bersih dan tidak ada goresan
2. Bersihkan kuvet dengan tisu sebeleum dimasukkan ke dalam cell holder
3. Sampel yang diisikan dalam kuvet jangan sampai melebihi garis segitiga pada kuvet
4. Jangan menumpahkan sampel di dalam spektrofotometer
Pengaturan gelombang cahaya yang digunakan
1. Pilih menu wavelenght
2. Isikan panjang gelombang yang diinginkan
3. Tekan enter

STANDARD OPERATING PROCEDURE (SOP)

SPECTROPHOTOMETER GENESIS 10S UV-Vis

1. Make sure the device is connected to electrical source

2. Press the on/off button located behind the device
3. Insert the sample-loaded cuvette into cell holder and blank-loaded cuvette into B-
marked cell holder
4. Seal the cell holder
5. Press B button on the button board
6. Press the “ ” button under the measure blank sign
7. Wait until “0.000 A” number pop out on the monitor screen
8. Press the “test” button until several menu pop out on the screen
9. Measure the wavelength on the wavelength menu, according to the desirable
wavelength
10. Set the number of sample used in “number of sample” menu.
11. Press the “test” button
12. And then, press the “run test” button.
13. Wait for the result to appear on the screen
14. Note and record the result then press the “esc” until “0,000 A” appear on the monitor
screen
15. Open the cell holder seal then remove the sample-loaded and blank-loaded cuvette.
16. Close the cell holder seal
17. Press the “on/off “ button behind the spectrophotometer
18. Disconnect the spectrophotometer from the electrical source
Few things that is important to note on the usage of Spectrophotometer
1. Make sure the cuvette is clean and no scratches before use
2. Clean the cuvette with tissue before inserted into cell holder
3. Do not overload the cuvette with sample, pay attention to the triangle-line limit on the
cuvette
4. Do not spill the sample inside the Spectrophotometer
How to adjust the wavelength
1. Choose the “wavelength” menu
2. Insert the desirable wavelength
3. Press “enter”

• Sambungkan pada sumber listrik, dan tekan tombol

on/off dibelakang spektrofotometer.

• Masukkan sample pada kuvet pada cell holder dan

sampel blanko pada cell B, tutup cell holder.

• Tekan tombol B pada papan tombol, lalu pada

papan tombol tekan tombol dan tunggu hingga
3 muncul 0.000 A pada papan layar.

• Tekan tombol test, atur panjang gelombang sesuai

dengan yang diinginkan, atur jumlah sampel, tekan
4 tombol test, tunggu hasil muncul.

• Catat hasil, kemudian tekan tombol esc hingga

muncul 0.000 A pada layar, buka cell holder dan
5 keluarkan kuvet serta blanko yang berisi sampel.

• Tutup kembali cell holder, tekan tombol on/off

untuk mematikan spektro, putuskan hubungan
6 dengan sumber listrik.

• Connect to the power source, and press the on / off button

behind the spectrophotometer.

• Insert sample-loaded cuvette into cell holder and blank-

loaded cuvette in cell B, close cell holder.

• Press the "B" button on the keyboard, then on the

keyboard press the button and wait for 0,000 A to
3 appear on the display screen.

• Press the test button, adjust the wavelength as desired, set

the number of samples, press the test button, wait for the
4 results to appear.

• Record the results, then press the esc key until 0.000 A
appears on the screen, open the cell holder and remove the
5 cuvette containing the blank and the sample.

• Close the cell holder again, press the on / off button to turn
off the spectro, disconnect it from the electrical source.
