ABSTRACT
BIOETHANOL PRODUCTION FROM SUGARCANE BASS (Saccharum
officinarum Linn.) USING Neurospora sp. IN SOLID-STATE CULTURE
MEDIA CULTURE)Ethanol can be produced from the bioconversion of
lignocellulosic biomass by Neurospora sp. through the process of
saccharification and fermentation simultaneously using solid-state or
submerged cultures. The focus of this study was to examine the potential for
the cultivation of Neurospora sp. using bagasse as a source of carbon and
nutrients, as well as a solid culture medium in the production of bioethanol.
In this study, the effect of the amount of inoculum used on
growth of Neurospora sp. and bioethanol production were studied. The ratio
of inoculum and bagasse (number of cells/weight of bagasse) which was
varied in this study was the ratio 1 (1.75 x 107); ratio 2 (2.5 x 107); and a
ratio of 3 (3.77 x 107) cells/g bagasse. Inoculum was prepared in liquid
Potato Dextrose Broth (PDB) medium using 20 g/L glucose as the sole
carbon source.
The specific growth rate of inoculum observed at ambient temperature and
initial pH of 5.3 was 0.0421 per hour with a cell doubling time of 16.5 hours.
Growth of Neurospora sp. on solid culture medium was only observed
qualitatively. Growth of Neurospora sp. at the ratio of inoculum and bagasse
3.77 x 107 cells/g bagasse looks better than the other two variations.
Meanwhile, the highest bioethanol production was produced at the inoculum
and bagasse ratio of 1.75 x 107 cells/g bagasse with a product yield of 13.6%
on the 4th day of fermentation. Thus, bagasse has the potential as a source of
nutrients as well as a solid culture medium in the bioethanol production
process.
