Saudara diminta untuk membuat media : tuliskan mulai dari perhitungan hingga

siap dipakai
1. Terdapat 5 sampel air yang akan dilakukan uji convirmatif, jika setiap sampel
membutuhkan 9 buah tabung media BGLB masing-masing berisi 10 ml. Bila
diketahui pada media tertulis :
Preparation : Suspend 40 g in 1 litre of demin. Water. Fill into tubes
fitted with fermentation tubes, autoclave 15 min at 121 0C. pH: 7,2
0,2 at 250C
2. Setiap hari laboratorium rata-rata menerima 6 sampel usap tenggorok yang
harus ditanam pada media blood agar, buatlah media tersebut untuk
kebutuhan 2 minggu [laboratorium libur setiap sabtu dan minggu]. Pada
media tertera :
Direction : Suspend 40 g of the powder in 1 L of purified water. Mix
thoroughly, heat with frequent agitation and boil 1 min. to completely
dissolve the powder. Autoclave at 121 0C for 15 min. to prepare blood agar
aseptically add 5% sterile defibrinated blood to the medium cooled to 45-50
0
C mix well pH: 6,8 0,2 at 250C
3. Untuk uji biokimia dibutuhkan 35 tabung media TSIA, bila disetiap tabung
diisi dengan 3 ml media. Pada botol media tertulis:
Direction : Suspend 65 g of the powder in 1 L of purified water. Mix
thoroughly, heat with frequent agitation and boil 1 min. to completely
dissolve the powder. Dispense into tubes and autoclave at 121 0C for 15 min.
cool in a slanted position so that deep butts are formed. pH: 7,4 0,2
4. Pada uji fermentasi dibutuhkan 40 tabung gula Glukosa, 25 tabung gula
Laktosa dan 50 tabung gula Manitol, indicator yang digunakan adalah Phenol
red dengan konsentrasi 4 %.
Resep buffer pepton : Suspend 25,5 gr in 1 litre of demin. Water, if desired
dispense into smaller vessels, autoclave 1210C for 15 min. pH: 7,0 0,2
5. Akan dilakukan isolasi terhadap 25 sampel feses pada media SS-Agar,
disetiap petri diisi dengan 20 ml media. Pada kemasan media tertulis :
Completely suspend 60g in 1 litre of demineralized water by heating in a
boiling water bath or in current of steam. Do not autoclaved, cool rapidly and
pour plates to give a thick layer of medium. pH: 7,0 0,2
