b. Antibodi Sekunder Setelah diinkubasi bersama antibodi primer, kertas mebran dibilas terlebih
dahulu barulah diinkubasi dengan antibodi sekunder. Antibodi sekunder adalah antibodi yang
spesifik untuk suatu spesies pada antibodi primer. Misalnya, anti-tikus hanya akan berikatan pada
antibodi primer yang berasal dari tikus. Antibodi sekunder biasanya berikatan dengan enzim
reporter seperti alkaline fosfatase atau horseradish peroxidase. Antibodi sekunder ini kemudian
akan menguatkan sinyal yang dihasilkan oleh antibodi primer. Sekarang, proses deteksi dapat
dilakukan dengan satu langkah saja, yaitu dengan menggunakan antibodi yang dapat mengenali
protein yang diinginkan sekaligus memiliki label yang mudah dideteksi 8. Analisis a. Colorimetric
detection Metode ini digunakan bila substrat dapat bereaksi dengan reporter enzyme sehingga
dapat mewarnai membran nitorselulosa b. Chemiluminescent Metode ini digunakan bila substrat
merupakan molekul yang bila bereaksi dengan antibodi sekunder atu dengan reporter enzyme akan
teriluminasi. Hasilnya kemudian diukur dengan densitometri untuk mengetahui jumlah protein yang
terwarnai. Teknik terbarunya yang paking canggih disebut Enhanced Cheiluminescent (ECL). Teknik
inilah yang paling banyak digunakan sekarang. c. Radioactive detection Metode ini menggunakan X-
ray yang bila mengenai label akan menciptakan region gelap. Namun metode ini sangat maha dan
beresiko tinggi terhadap kesehatan. d. Fluorescent detection Pelacak yang mmepunyai label yang
dapat mengalami fluorosensi lalu kemudian dideteksi oleh fotosensor seperti kamera CCD yang
menangkap image digital dari western blot. Hasil kemudian dapat dianalisi secara kuantitaif maupun
kualitatif Metode ini juga merupakan salah satu metode yang paling sering digunakan karena sangat
sensitif. PRINSIP KERJA WESTERN BLOT ANALYSIS Prinsip kerja Western Blot yaitu pertama kali
dilakukan analisis dengan SDS-PAGE karena protein mempunyai berat molekul yang beragam,
sehingga setelah dilakukan SDSPAGE, protein dengan berat molekul yang berbeda akan terpisah
pada area gel. Kemudian dilanjutkan dengan mentransfer protein tersebut dari Polyacrilamide Gel ke
membrane nitroselulose dan selanjutnya dilabel dengan antibodi. Hasil ikatan antara protein antigen
dan antibodi tersebut kemudian diwarnai dengan menggunakan pewarnaan yang diinginkan, seperti
pewarnaan dengan commasie blue (Rantam, 2003). Gambar 1. Rincian langkah-langkah yang terlibat

First things first. Obtain a protein sample you want to analyze, such as cell samples. Lyse the cells to
release protein contents. Run these on a gel which separates proteins on the basis of size. Then
transfer these gel proteins onto a membrane using electricity. This membrane can then be used to
probe for proteins of interest using a primary antibody. What You Need to Western Blot: A Protein
Sample A Good Antibody to Detect your Protein of Interest Western blot relies on the primary
antibody to detect this protein from the thousands of proteins on your membrane and previously on
your gel! (a cell can contain 30,000 different proteins - and these same proteins can even be altered
giving you over 300,000 different proteins!). Using an antibody recognizes your primary antibody (a
secondary antibody) you build up a protein-antibodyantibody sandwich! The secondary antibody has
a horse radish peroxidase enzyme which converts a luminol substrate to a light releasing substance!
This light is detected as a spot on film. From this spot you can determine how much protein is there
relative to other spots, or the size of the protein relative to a size marker that is run also on the gel.
Diagram 2 shows a western blot example gel. Lane 1 is a protein size marker ladder which shows
different known sizes of proteins, this can be purchased commercially and the sizes of all the spots
are given in a pamphlet. Lane 3 is a cancer sample and lane 5 is a normal sample. As you can see the
protein in lane 3 has a higher expression than the cancer sample in lane 5, which is interesting. Also,
the protein spots in lanes 3 and 5 are the same size as the 2nd spot in the size ladder from lane 1.
We can then look at the known protein size from our brochure which we received with the ladder.
We then determine that the size of the protein is 80 kDa. Our protein of interest is also 80 kDa. So
we know that the western blot worked and that the protein is highly expressed in a cancer sample!
