g/mL FEA meningkat sel Vero hidup hingga 10,24% sedangkan silymarin
sebesar 12,54%. CCl4 dosis 2 mL/kg BB diberikan pada hari ke-4 dan ke-8
mengakibatkan tekanan pertambahan berat badan (BB) tikus SD jantan,
menimbulkan kerusakan hepatosit seperti degenarasi hidrofik, steatosis, dan
akumulasi glikogen, serta ditandai dengan meningkatnya serum alanin
transaminase (ALT), aspartat transaminase (AST), dan conjugated bilirubin (CB).
Pemberian senyawa AO (FEA M. subpeltata dosis 50, 100, 150 mg/kg BB; 100
mg/kg BB silymarin; dan butil hidroksi anisol/BHA dosis 10 mg/kg BB) selama 10
hari mampu mencegah tekanan pertambahan BB dan kerusakan liver akibat
pemberian CCl4. FEA dan silymarin dosis 100 mg/kg BB memiliki kemampuan
yang setara dalam mencegah penurunan BB sedangkan BHA 10 mg/kg BB
memiliki kemampuan yang paling rendah. Senyawa AO mampu mencegah dan
memperbaiki kerusakan hepatosit dari SO yang diakibatkan oleh metabolisme
CCl4. FEA M. subpeltata mampu mencegah lipid peroksidasi yang ditandai dengan
kadar malondialdehid (MDA) yang normal, meningkatkan protein total liver, dan
meningkatnya AO endogen liver (terutama SOD dan GSt) yang sebanding dengan
silymarin maupun BHA. AAO FEA M. subpeltata dalam mencegah SO baik in
vitro maupun in vivo melalui mekanisme: penangkapan radikal bebas, agen
pereduksi, pengkelat logam, pencegah peroksidasi lipid, dan aktivasi antioksidan
endogen. Tumbuhan obat yang digunakan oleh pengobat tradisonal merupakan data
base yang sangat penting untuk mengekplorasi senyawa antioksidan baru. Faksi etil
asetat dari M. subpeltata menunjukkan potensi yang baik untuk dimanfaatkan lebih
lanjut.

at the same dose of 150 g.mL-1 were able to reduce the ROS production of 50%
and 44% respectivally. The giving of FEA 50 g.mL-1 increased Vero cells alive
by 9.53% while silymarin 50 g.mL-1 was 8.09%. Compared to untreated cells,
EAF at a dose of 150 g.mL-1 increased the alive of Vero cells by 10.24% while
silymarin was 12.54%. CCl4 at doses of 2 mL.kg-1 BW by gavage administration
on the 4th and 8th days indicated an oppression on male SD rats' weight gain. Rats
receiving the CCl4 displayed hepatocyte damage such as hydropic degeneration,
steatosis, and accumulation of glycogen. It also markedly elevated serum alanine
transaminase (ALT), aspartate transaminase (AST), and conjugated bilirubin (CB).
A consecutive 10 day-administration of AOs (EAF M. subpeltata at doses of 50,
100, 150 mg.kg-1 BW; silymarin at a dose of 100 mg.kg-1 BW; and butyl hydroxy
anisol/BHA at a dose of 10 mg.kg-1 BW) were capable of discharging weight gain
oppression and alleviating of liver damage due to CCl4 treatment. EAF and
silymarin at a dose of 100 mg.kg-1 BW exhibited the same capacity to prevent
weight loss, while BHA 10 mg/kg BW resulted the lowest demonstration. AO
compounds can prevent and repair OS hepatocyte due to CCl4 intoxication. The
EAF of M. subpeltata decreased malondialdehyde (MDA) to a normal level an
indication of lipid peroxidation prevention, increase of total liver protein, and
enhance liver endogenous AO (especially SOD and GST) of SD rats compared to
those CCl4-treated rats which were equal to the effect of silymarin and BHA
administration. The AOA mechanisms in which FEA of M. subpeltata preventing
OS both in vitro and in vivo were through: free radical scavenging, reducing
agents, metal chelating, preventing lipid peroxidation, and activating of
endogenous antioxidants. Medicinal plants used by traditional healers are a very
important database for exploring new antioxidant compounds. The ethyl acetate
fraction of M. subpeltata showed potential AAO properties for further utilization.
