PENDAHULUAN
• Virus di alam bersifat unik, merupakan partikel terkecil yang
mampu melakukan replikasi.
• Dalam bentuk yang paling dasar, virus hanya terdiri dari
segmen kecil dari asam nukleat yang terbungkus oleh protein
sederhana
• Virus tidak memiliki sistem metabolisme sendiri, bersifat
obligat dengan menginvasi sel dan memparasit sistem
subseluler inang
• Virus dapat menginfeksi setiap organisme di alam
• Definisi : partikel yang mempunyai kemampuan menginfeksi
dan bereplikasi di dalam inang.
SIFAT-SIFAT VIRUS
• Virus adalah parasit intraseluler obligat
• Diameter : 20 nm-300 nm
• Hanya memiliki satu asam nukleat : DNA/RNA saja
• Struktur : terdiri dari kapsid yang melindungi asam
nukleat beberapa virus dilengkapi dengan
envelope
• Bermultiplikasi di dalam sel hidup, dengan
menggunakan sistem reproduksi yang dimiliki oleh
sel hospes
Terminologi Virologi
Protein subunit
Molekul protein tunggal yang
terlipat
Satu subunit protein
tersusun atas satu rantai
polipeptida yang dikode oleh
satu gen
molekul protein tunggal yang
diassembly dengan molekul
protein lain membentuk
kompleks protein
Terminologi Virologi
Subunit Struktural
• Unit penyusun capsid, dapat
tersusun oleh satu atau
banyak protein subunit yang
berbeda, biasanya
dinamakan protomer.
Batang
(Helical Virus)
Struktur Komplek
(Complex Structure)
Virus Ikosahedral (Icosahedral virus)
• Ikosahedron tersusun atas 20
permukaan (masing masing
sigitiga sama sisi), 12 vertex
(puncak) dan 5 lipatan, tiga
lipatan dan dua lipatan aksis axia
simetri yang rasional.
• Vertek mempunyai 5 unit yang
bersebelahan (pentavalen) dan
yang lain mempunyai 6 unit yang
bersebelahan (hexavalent).
• Contoh : Adenovirus, Picornavirus
Campbell, Reece, Urry et all. Biology. Ed 8th
Ikosahedral virus
• Untuk membentuk
struktur ikosahedron
dari molekul protein
yang identik dibutuhkan
60 molekul
• Setiap segitiga sama sisi
disusun oleh tiga
molekul
Virus Heliks (Helical Virus)
• Subunit-subunit protein identik
(protomer) terasembling menjadi
kapsid melalui model heliks
mengelilingi asam nukleat secara
spiral
• Kapsid heliks dikarakterisasi Campbell, Reece, Urry et all. Biology.
Ed 8th
berdasarkan panjang, lebar dan pitch
heliks serta jumlah protomer setiap
heliks
• Contoh : virus tanaman mosaik
tembakau tersusun 1000 lebih
protein (sejenis)
Jawetz, Melnick dan Adelberg. Medical
Microbiology. Ed 24th.
Virus mosaik tembakau (TMV). Kapsid terdiri atas 49 subunit protomer dan terasembling dalam 3
putaran heliks, panjang setiap putaran heliks adalah 2,3 nm. Virus lengkap memiliki ukuran
p.300X d.18 nm dan diameter cylindrical core 8 nm
Virus Komplek (Complex Virus)
• Terutama virus bakteri
• Lebih komplek dari ikosahedral
dan virus helik
• Contoh : Bakteriofag T4
kapsid komplek (kepala
ikosahedral+aparatus ekor)
Ordo (-virales)
Subfamili (-virinae)
Genus (-virus)
pH Radiasi
• Umumnya pH 5,0 - 9,0 • UV, Sinar X dan partikel
• Virus rentan terhadap basa berenergi menginaktifkan
• Enterovirus resisten virus
terhadap asam
Reaksi Virus terhadap Agen Kimia dan Fisika
Eter Detergen
• Detergen noninonik :
• Setiap kelompok virus melarutkan lipid pada
bersifat sensitivitas amplop dan denaturasi
terhadap eter protein kapsid
• Detergen anionik : bersifat
solubel dgn amplop dan
menghancurkan polipeptida
kapsid
Reaksi Virus terhadap Agen Kimia dan Fisika
Formaldehid Antibiotik
• Sifat menginaktifasi genom • Antibiotik dan antibakteri
virus lain tidak efektif
• Bereaksi dengan asam • Telah ada antivirus
nukleat • Dibanding dengan bakteri
• Lebih mudah pada untai konsentrasi dibutuhkan
ganda lebih besar (Klorin)
• Alkohol, isopropanol, etanol
relatif tidak efektif

Replikasi Virus
• Replikasi terjadi di
nucleus, melibatkan
pembentukan suatu
utas (-)sense yang
bertindak sebagai
template bagi
dibuatnya utas
(+)sense.
Replikasi virus DNA
Class VII: Double-stranded DNA with RNA intermediate
(Hepadnaviruses):
• Kelompok virus ini memerlukan enzim reverse
transcriptase,tetapi tidak seperti
Retrovirus,pembentukan RNA intermediate terjadi
dalam partikel virus saat proses maturasi. Saat
menginfeksi sel inang baru, genom virus akan
memperbaiki gap yang ada dan melanjtkannya dengan
proses transkripsi.
• Agar replikasi genomberlangsung sukses, virus
mempresentasikan mRNAke sel inang dan selanjutnya
di translasi menjadi protein virusdengan beberapa
cara :
– d/s DNA viruses release their genome into the
nucleus and rely on the cellular machinery for
transcription.
– (+)sense RNA viruses have genomes which
serve directly as mRNA (except retroviruses!)
– (-)sense RNA viruses must carry with them
virus-coded enzymes for RNA-dependent
RNA replication, since uninfected cells do not
posses these enzymes.
Replikasi virus DNA
5. TRANSLASI
Translasi mRNAs mejdai protein
berlangsung di sitoplasma di
ribosom dan dihasilkan protein
struktural dan protein kontrol dari
virus.
Replikasi virus DNA
6. REPLIKASI
• Replikasi asam inti (NA) virus terjadi
dinucleus, kecuali iridovirus dan
poxvirus. In these, the NA replicates in
the cytoplasm so that DNA synthesis and
viral protein synthesis occur in a
common cytoplasmic factory.
• Cellular DNA polymerase is involved in
replication of viral NA, except for the
larger DNA viruses which code for their
own DNA polymerase Control of viral
replication is achieved by tight regulation
of gene expression. The methods used
depend on nature of the virus
genome/replication strategy, e.g:
Segmented genomes are usually
transcribed to produce monocistronic
mRNAs. One advantage of
monocistronic mRNAs is that various
7. ASSEMBLY/PERAKITAN
Involves the assembly of all the components
necessary for the formation of the mature
virion at a particular site in the cell. During
this process, the basic structure of the virus
is formed.
