Mempelajari mekanisme kerja
obat-obat antihiperglikemia
dengan metode pembebanan
glukosa
 Hiperglikemia adalah kondisi
ketika kadar gula didalam
darah melebihi batas normal
yang jika tidak diobati dapat
menyebabkan komplikasi,
terutama pada penderita
diabetes.
Diabetes mellitus (DM) adalah penyakit kronis yang disebabkan oleh
ketidakmampuan tubuh untuk memroduksi hormone insulin / karena
penggunaan yang tidak efektif dari insulin. Hal ini ditandai dengan
tingginya kadar glukosa dalam darah. Menurut klasifikasinya (DM)
dibedakan menjadi:

Tipe 1 (DMT 1) adalah insufisiensi absolut insulin

01 (tidak adanya produksi insulin sama sekali

02 Tipe 2 (DMT 2) disebabkan tidak cukup

dan tidak efektifnya kerja insulin

03 Diabetes Gestasional, yang muncul saat

hamil

04 Gangguan toleransi glukosa (GTG), kadar glukosa

antara normal dan diabetes, dapat menjadi
diabetes atau menjadi normal atau tetap tidak
berubah
Berdasarkan mekanisme kerjanya, obat hipoglikemik dapat dibagi
menjadi 5 golongan, yaitu:

Gol. Sulfoniluera Gol. Biguanid Gol. Analog Meglitinid

 01 Mekanisme Kerja Obat Gol.Sulfonilurea
(Glipenklamid)

Bekerja dengan cara merangsang sekresi

insulin dipankreas sehingga hanya efektif bila
sel beta pankreas masih dapat berproduksi.
Sulfonilurea menstimulasi pelepasan insulin
daei sel beta pancreas.
 02 Mekanisme Kerja Obat Gol.Biguanid (Metformin)

Bekerja dengan cara menghambat gluconeogenesis dan

meningkatkan penggunaan glukosa di jaringan.
Metformin menurunkan kadar glukosid terutama
dengan menurunkan produksi glukosa hepatic dan
dengan meningkatkan kerja insulin di otot dan di lemak.
Mekanisme Metformin mengurangi produksi glukosa
hepatic ini bersifat kontroversial, tetapi sebagian besar
mendukung efek penurunan gluconeogenesis.

Analog Meglitinid:
Mengikat reseptor sulfonylurea
dan menutup ATP-sensitive
potassium chanel.
Mekanisme Kerja Obat Gol.
Thiazolidindion:
Meningkatkan sensitivitas
jaringan perifer terhadap
insulin.
Mekanisme Kerja Obat Gol.
Penghambat
Alphaglukosidase:
Menghambat
alphaglukosidase yang
mengubah di/polisakarida
menjadi monosakarida,
sehingga memperlambat dan
menghambat penyerapan
karbohidrat.
