when coupled to a carrier molecule.
Hapten hanya akan menjadi imunogenik jika
diikat oleh molekul yg besar (disebut carrier)
yang dikenal sistem imun dan baru dapat
menstimulasi pembentukan Antibodi.

Pembagian Ag menurut Epitop
1. Unideterminan,univalen : hanya 1 jenis
determinan/epitop pd 1 molekul. Contoh : hapten
2. Unideterminan, multivalen : hanya 1 jenis epitop
ttp 2 atau lebih epitop tsb ditemukan pada 1 molekul.
Contoh : polisakarida
3. Multideterminan, univalen : byk epitop bermacam-
macam, tp hanya 1 dari tiap macamnya. Contoh :
protein
4. Multideterminan, multivalen : byk macam epitop &
byk dari tiap macam pd 1 molekul. Contoh :
kompleks kimiawi dan BM yang tinggi

Pembagian Ag menurut spesifisitas
1. Heteroantigen : dimiliki oleh banyak spesies
2. Xenoantigen : dimiliki oleh spesies tertentu
3. Alloantigen (Isoantigen) : spesifik untuk
individu dalam satu spesies
4. Antigen organ spesifik : hanya dimiliki organ
tertentu
5. Autoantigen : milik tubuh sendiri walau
merupakan unsur jaringan normal tetapi
menjadi target respon imun (pd penyakit
autoimun)

Immunogen yang poten biasanya memiliki
BM>10.000 (Cth: protein dg BM yg besar)
Molekul dengan BM<10.000 bersifat
kurang immunogenik. Asam amino atau
monosakarida biasanya tidak bersifat
antigenik
Kecuali: hapten (protein dg BM kecil), tp jk
berikatan dg molekul carrier Antigenik

Hal ini karena kebanyakan jenis Ag
(spt: T-dependent Ag), jika akan
membentuk imun respon, terlebih
dahulu harus dikenal, kemudian
diproses, baru dipresentasikan
kepada T-helper oleh APC (Antigen
Presenting Cell)

Sifat Kimia Imunogen
1. Protein :
• Mayoritas imunogen protein
• Umumnya epitop multideterminan, univalen.
• Dapat berupa protein murni atau kombinasi
spt :
Lipoprotein (terdapat pada membran sel),
Glikoprotein (zat pembentuk gol darah A,B,O)
reaksi transfusi & pembentuk Ag Rhesus
isoimunisasi pada kehamilan.

2. Polisakarida/Hidrat arang:
Polisakarida murni (cth: kapsul
Pneumococcus menghasilkan Ab
Ab anti Pneumococcus),
Lipopolisakarida (cth: ddg sel bakteri
Gram neg endotoksin terhadap
V. cholera)

3. Asam Nukleat : biasanya kurang bersifat
imunogen kecuali jika berikatan dengan
carrier protein. DNA dalam bentuk helix
biasanya tidak imunogenik. Tetapi
respon imun terhadap DNA dapat terjadi
pada penderita dengan SLE (Systemic
Lupus Erythematosus)

Jenis-jenis Antigen
1. T-independent Ag : Ag yang dapat
langsung menstimulasi sel B
menghasilkan Ab tanpa perlu
bantuan sel T. Termasuk dalam
T-independent Ag: lipopolisakarida,
dekstran, flagelin polimerik bakteri.

T-independent Ag
Ciri khas Ag ini :memiliki epitop yang sama
dan berulang. (Gambar 1)
Berupa molekul besar polimer yang dipecah di
dalam tubuh secara perlahan-lahan.
Sifat aktivasi sel B secara poliklonal : Ag ini
dapat mengaktivasi sel B spesifik untuk jenis
Ag yang lain. Berdasarkan kemampuan ini,
dibagi 2 subtipe:
Tipe 1 poliklonal aktivator
Tipe 2 tidak bersifat aktivator.

Antigen (Ag)
substance that provokes an immune response in
specific lymphocytes
perceived as foreign, not a normal constituent of
the body
Foreign cells & large complex molecules over
10,000 MW are most antigenic
Foreign molecules <1,000 MW (haptens) are not
antigenic unless attached to a larger carrier
Antigenic determinant, epitope – small molecular
group that is recognized by lymphocytes. An
antigen has many epitopes.

Special categories of antigens
Autoantigens – molecules on self tissues for
which tolerance is inadequate
Alloantigens – cell surface markers of one
individual that are antigens to another of that
same species
Heterophilic antigens – molecules from unrelated
species that bear similar antigenic determinants
Superantigens – potent T cell stimulators,
provoke an overwhelming response
Allergen – antigen that provokes allergy

T-cell dependent antigens must be processed by
phagocytes called antigen presenting cells (APC).
APC alters the antigen and attaches it to its MHC
receptor.
Antigen presentation involves a direct collaboration
among an APC, T helper cell and an Ag-specific B
or T cell.
Interleukin -1 is secreted by APC to activate T H cells
Interleukin-2 is produced by TH to activate B & other
T cells
