ABSTRAK
Diabetes mellitus (DM) merupakan penyakit gangguan metabolisme kronis,ditandai
dengan tingginya kadar glukosa dalam darah (hiperglikemia) yang disebabkan karena penurunan
sekresi insulin. Pengobatan yang penting untuk Diabetes Mellitus (DM) tipe 2 adalah
mengendalikan kadar glukosa darah. Enzim alfa amylase di dalam tubuh berperan memecah
karbohidrat menjadi amylum. Pengendalian enzim amylase tersebut sangat diperlukan pada kasus
diabetes. Salah satu tanaman yang memiliki efek hiperglikemia adalah daun tin (Ficus carica L).
Tujuan dari penelitian ini yaitu untuk mengetahui aktivitas penghambatan enzim alfa amylase oleh
ekstrak etanol dan fraksi n-heksan, dan fraksi etil asetat dari daun tin (Ficus carica L.) secara in
vitro menggunakan metode spektrofotometri UV-Vis dengan inhibitor standar akarbose sebagai
pembanding. Akarbose dapat menunda hidrolisis karbohidrat dan disakarisa, absorbsi gula dan
menghambat metabolisme sukrosa menjadi glukosa dan fruktosa. Dari hasil pengujian yang telah
dilakukan, diketahui bahwa daun tin (Ficus carica L) mengandung senyawa metabolit sekunder
meliputi flavonoid, alkaloid, tannin dan steroid atau triterpenoid. Sementara itu hasil pengujian
penghambatan enzim alfa amylase oleh ekstrak etanol dan fraksi diperoleh nilai IC50 berturut-turut
sebesar 138,96 µg/mL, 117,78 µg/mL,dan 147,50 µg/mL. Berdasarkan hasil tersebut, dapat
disimpulkan bahwa ekstrak etanol, fraksi n-heksan dan fraksi etil asetat daun tin (Ficus carica L)
memiliki aktivitas penghambatan enzim alfa amylase dengan kategori sedang.

ABSTRACT
Diabetes mellitus (DM) is a chronic metabolic disorder, characterized by high levels of
glucose in the blood (hyperglycemia) caused by decreased secretion insulin.An important treatment
for type 2 diabetes is controlling blood glucose levels. Alpha amylase enzyme in the body plays a
role in breaking down carbohydrates into strach. Control of the amylase enzyme is needed in
diabetes cases. One of the plants that have a hyperglycemic effect is Tin (Ficus carica L). This
study aimed to determine the inhibitory activity of the alpha amylase enzyme by ethanol extract and
n-hexane, ethyl acetate fractions from Tin (Ficus carica L) leaves in vitro using UV-Vis
spectrophotometri method with a standard inhibitor of acarbose as a comparison. Acarbose can
delay the hydrolysis of carbohydrates and disaccharides, absorption of sugar and inhibit the
metabolism of sucrose into glucose and fructose. The result showed that Tin leaves contained
secondary metabolites including flavonoids, alkaloids, tannins, and steroids or triterpenoids.
Meanwhile, the results of the alpha amylase inhibition test by ethanol extract and fractions
obtained IC50 values of, 138,96 µg/mL, 117,78 µg/mL,dan 147,50 µg/mL. Based on these results, it
can be concluded that the ethanol extract and n-hexane, ethyl acetate fractions from Tin leaves
have moderate activity in inhibiting the alpha amylase enzyme.
