II. TUJUAN
Mahasiswa mampu mengoperasikan software Toxtree dan memanfaatkannya untuk
meprediksi parameter toksisitas beberapa senyawa kimia.
III.
DASAR TEORI
Toksikologi adalah pengetahuan tentang efek racun dari obat terhadap tubuh
dan sebenarnya termasuk pula dalam kelompok farmakodinamika, karena
efek terapeutis obat berhubungan erat dengan efek toksisnya. Pada
hakikatnya setiap obat dalam dosis yang cukup tinggi dapat bekerja sebagai
racun dan merusak organisme (Sola dosis facit venenum: hanya dosis
membuat
racun, Paracelsus).
Pada
umumnya,
hebatnya
reaksi
toksis
secukupnya. Obat teratogen adalah obat pada dosis terapeutis untuk ibu
hamil dapat menyebabkan cacat pada janin, seperti focomelia. Toksoid atau
anatoksin adalah suatu toksin yang telah diubah strukturnya, sehingga tidak
terjadi toksik lagi. Sifat antingennya tidak dihilangkan, yakni kemampuan
untuk menstimulasi pembentukan antibodi (Tjay & Rahardja, 2002).
Sebelum percobaan toksikologi dilakukan sebaiknya telah ada data
mengenai identifikasi, sifat obat dan rencana penggunaannya. Data ini dapat
dipakai untuk mengarahkan percobaan toksisitas yang akan dilakukan. Hal
ini memerlukan judgement seorang yang berpengalaman dalam bidang ini.
Respon berbagai hewan coba terhadap uji toksisitas sangat berbeda, tetapi
hewan coba yang lazim digunakan ialah salah satu strain tikus putih. Kadangkadang digunakan mencit dan satu dua spesies yang lebih besar seperti
anjing, babi atau kera. Batas dosis harus dipilih sedemikian rupa sehingga
dapat memperoleh suatu kurva dosis respons yang dapat berwujud respons
bertahap (misalnya mengukur lamanya waktu tidur) atau suatu respons
kuantal (misalnya mati). Biasanya digunkan 4-6 kelompok terdiri dari
sedikitnya 4 ekor tikus. Cara pemberian obat harus dipilih sesuai dengan
yang digunAkan di klinik. Jadi untuk obat yang akan dipakai sebagai obat
suntik perlu diuji dengan cara parenteral dan obat yang digunakan sebagai
salep terutama harus diuji terhadap kulit (Katzung, 2001).
Ada beberapa kemungkinan untuk menggolongkan toksikologi yang
dapat dibagi menjadi dua yaitu efek toksik akut yang langsung berhubungan
dengan pengambilan zat toksik sedangkan efek toksik kronis yang pada
umumnya zat dalam jumlah sedikit diterima tubuh dalam jangka waktu yang
lama sehingga akan terkumulasi mencapai konsentrasi toksik dan dengan
demikian menyebabkan terjadinya gejala keracunan (Mutschler, 1991).
Efek samping toksik bergantung kepada dosis dan sfesifik bagi obat.
Sepanjang di berikan dosis yang cukup tinggi, Efek samping toksis terjadi
pada setiap orang karena toleransi perorangan terhadap suatu obat sangat
beragam, selalu terdapat kemungkinan bahwa akibat dosis yang dapat di
terima kebanyakan pasien, pada beberapa penderita terjadi efek samping.
Penyebab pokok jenis variasi biologi kini mirip dengan perbedaan kerja
dalam percobaan hewan mungkin adalah perbedan perbedaan akibat
konstitusi atau genetic dalam absorpsi, distribusi, biotransformasi dan /atau
eliminasi, yaitu dalam farmakokinetika bahan obat, serta dalam kerapatan
reseptor termasuk ditribusi reseptor. Disamping perbedaan penerimaan obat

meprediksi toksisitas akuatik dan chemosphere. Berdasarkan aturan verhaar,
Antracene masuk ke dalam kelas 1 (narcosis/baseline toxicity), hal ini karena
antrasen hanya mengandung unsur C dan H.

Pada parameter Benigni (Mutagenesis dan Karsinogenisis) senyawa Antracene
termasuk dalam genotoksin pada karsinogen yang menyebabkan kerusakan
genetik ireversibel atau mutasi dengan mengikat DNA.Genotoxins termasuk
agen kimia seperti N-nitroso-N-methylurea (NMU) atau agen non-kimia
seperti sinar ultraviolet dan radiasi pengion.Virus tertentu juga dapat bertindak
sebagai karsinogen dengan berinteraksi dengan DNA.
d. Structur Allert for the in vivo micronucleus assay in rodents
Pada struktur Antracene terdapat ikatan Polycyclic Aromatic Hydrocarbons,
dengan tiga atau lebih cincin. Tidak termasuk senyawa heterosiklik sehingga
antrasen digolongkan dalam golongan yang berperan dalam struktur alert
sebagai salah satu tanda positif untuk uji nukleus.

mengandung gugus fungsional yang berhubungan dengan peningkatan
toksisitas, masuk pada Q3 dimana tidak mengandung unsure-unsur lain selain
C, H, O, N, dan S, dimana Q5 menunjukkan aflatoxin mengandung
hidrokarbon alifatik seningga termasuk dalam kelas 1

normal, tidak ada kelompok fungsional yang terkait dengan tikngkat toksisitas,
tidak ada sebagian elemen yang tidak tercantum pada Q3 dan hanya terjadi Na,
K, Ca, Mg atau N garam dari asam karboksilat, atau sulfat atau hidroklorida
dari amina, atau sulfonat Na, K, atau Ca, sulphamate atau sulfat, dimana
semua elemen tersebut tidak terdapat Kloroform.
b. Verhaar
Aturan ini digunakan untuk mengklasifikasikan senyawa yang dapat menjadi
polutan lingkungan yaitu dengan melihat hubungan struktur aktivitas untuk
meprediksi toksisitas akuatik dan chemosphere. Kloroform masuk ke dalam
