BAGAIMANA CARA MEMVALIDASI?
Ayo memvalidasi metode !
• Siapa?
• Laboratorium yang sedang develop sebuah metode bertanggung
jawab atas benarnya validasi metode tersebut.
• Metode tidak baku, metode yang didesain/dikembangkan laboratorium,
metode yang digunakan di luar lingkup yang dimaksud, penegasan
serta modifikasi dari metode baku.
• Laboratorium menggunakan metode uji (official) bertanggungjawab
untuk mengecek bahwa metode yang tervalidasi tersebut benar
dalam penggunaannya.
• Apa?
• Praktisi laboratorium harus menentukan sejumlah parameter yang
diperlukan selama proses validasi, dan ini tergantung tingkat
validasi yang diperlukan (the degree of validation of the method)

• Seseorang harus merencanakan sebuah eksperimen denga tujuan
mendapatkan data statistik dari parameter pengukuran (sebuah
rencana matang dapat menghemat waktu dengan karakteristik
• Hasil?
• Seseorang harus dapat mengambil informasi yang berguna dari
parameter pengukuran (menggunakan statistik dan excel)
• Seseorang harus mengambil kesimpulan atas hasil yang didapat
dalam sebuah laporan yang mendetail.
Parameter utama yang harus divalidasi
• Ruang lingkup aplikasi
• Menjelaskan (keadaan analit), bagaimana metode uji menunjukkan
kesesuaian pengukuran/pengujian analit dalam suatu matrik dan
pada level konsentrasi yang diharapkan.
• Rentang kerja (ukur)
• Menjelaskan rentang konsentrasi dimana metode uji diaplikasikan
yang dinyatakan dalam presisi, akurasi (trueness) dan linieritas.

• The Limit of Detection (LoD) is the lowest concentration
that can be detected with an acceptable confidence.
• The Limit of Quantification (LoQ) is the lowest
concentration that can be quantified with an acceptable
precision and accuracy.
• Decision Limit (CCα), Detection Capability (CCβ)
• The decision limit (CCα) is the lowest concentration at which one
can be sure that the analyte is present (equal to or greater than
LoD).
• The detection capability (CCβ) is the smallest content of the
substance that can be detected, identified and/or quantified in a
sample (equal to or greater than LoQ).

• Kebenaran (Trueness)
• Kebenaran (Trueness) suatu metode adalah ukuran sejauh mana
kedekatan hasil pengukuran dengan nilai sebenarnya.
• Ketepatan (Precision)
• Ketepatan menggambarkan tingkat kedekatan antara hasil uji
independen yang diperoleh di bawah kondisi yang sama yang
dinyatakan dalam repeatability (pengulangan dengan perbedaan
waktu yang relatif singkat antar pengukuran independen) dan
reproducibility (pengulangan dengan perbedaan waktu relatif lama
antar pengukuran independen).
Parameter utama yang harus divalidasi
• Robustness (ruggedness)
• Test Robustness adalah pengukuran kapasitas metode uji untuk
tetap tidak terpengaruh oleh perubahan kecil dari lingkungan
dan/atau kondisi operasional. Perlakuan tersebut sengaja
dilakukan untuk melihat perubahan karakteristik parameter metode
uji.
• Ketidakpastian Pengukuran (Measurement Uncertainty)
• Ketidakpastian adalah suatu parameter yang terasosiasi dengan
hasil pengujian/pengukuran yang mencerminkan ketersebaran
nilai-nilainya yang layak dimiliki pada benda yang diuji/ukur.

BAGAIMANA MENGENALI METODE
YANG TELAH DIVALIDASI?
Bagaimana anda mengenali metode yang
telah divalidasi
• Evaluate method performance parameters
• Are target values achieved?
• YES – Method is fit-for-purpose in its field of application meaning
that Method is validated.
• NO – More development required and method has then to be
revalidated.
Kesimpulan
• Validasi Metode sangatlah dibutuhkan! Walaupun akan
menambah biaya yang harus dikeluarkan namun tanpa
validasi metode, analisis bagaikan TANPA nilai.
• There is not one way of performing validation: tingkat dan
parameter validasi akan sangat bergantung pada
keadaan (kadangkala verifikasi saja cukup). Dalam
keadaan tertentu, sebuah parameter PENTING bagi
situasi khusus, dan tidak significant pada situasi yang
lain: use common sense!
• Validasi Metode adalah sebuah proses yang harus
direncanakan dan berdasarkan ‘reliable data’.

Experimental design: method
• Identifikasi 3 parameter (A, B, C) untuk dipelajari
• Identifikasi keadaan normal (level 1) dan alternatif nilai-
nilai (level 2) untuk setiap masing-masing parameter.
• Lakukan 4 eksperimen yang dijelaskan berikut ini:
Example
• Suatu residu antibiotik diekstrak dari susu dengan
menggunakan pelarut.
• Hasil ekstrak dituangkan ke dalam suatu cartridge silika
dan dicuci dengan air.
• Antibiotik kemudian dielusi dengan acetonitrile dalam air
(5% v/v) dan dikuantifikasi dengan RP-HPLC
• Test robustness ini bertujuan untuk mengevaluasi step
clean-up
• Test robustness digunakan pada sampel susu yang
di“spike” dengan antibiotik pada konsentrasi sekitar 1,2
µg/l

tentukan mean
• Check masing-masing kelengkapan
• Number of trials for each experiment
• Standard deviation of repeatibility
• Number of replicates used for repeatibility calculation

• Kontrol parameter KUNCI dengan cara sedemikian rupa
sehingga tidak ada problem karena variasi parameter.
• Inilah cara yang disebut ROBUST !

Pembuatan Control Chart
• Control chart menampilkan nilai rata-rata untuk
mendisplay kontrol mutu dari suatu proses analisis.
• Bila data standard deviasi dan mean dari sampe (QC
sample) dilakukan secara routine control chart sangat
disarankan
• Grafik Control Chart menggambarkan hubungan mutu
hasil test dari waktu ke waktu atau sequence pengujian
dengan batasan nilai (specs) sesuai dengan kontrol
statistik yang telah ditetapkan.
Pembuatan Control Chart
Tahapan rasional untuk membuat Control Chart:
1. Identifikasi elemen-elemen penting yang akan menyusun
CC.
2. Tetapkan tujuan.
3. Pilih karakteristik yang akan diukur.
4. Tetapkan how, where, and when karakteristik yang akan
diukur.
5. Tetapkan besarnya set data.
6. Tetapkan prosedur untuk mendapatkan random sampel.
7. Tentukan estimasi rerata proses dan SD untuk jangka waktu
yang lama.
8. Buatlah lay out garis tengah (mean), garis peringatan, dan
batas tindakan (action limit).
Control Chart
Types of control chart
1. Precision controls charts based upon standard
solutions:
• Standard control should be run once every
2. Precision controls charts based upon duplicate
analyses:
• Test samplw examined should be within a narrow concentration
range;
• SD usually depends upon concentration and standard deviation of
differences between duplicate analyses also depends upon
concentration;
• At least 10 duplicate test sample (20 analyses) should be analyze
to construct a CC.
• The differences between the paired results are plotted with zero as
the expected value.
Batasan Control Chart
Types of control chart (lanjutan)
• Precision control charts based upon “Spiking” recovery
tests
• This CC are intended to determine bias in analyses, and provide
evidence of the lack of statistical control of appropriate part of the
measurement process.;
• Spike recovery test should only be applied in cases where RSD is
know to be small and the equivalent concentration of the spike
added lies between one and 10 times the concentration of the
analyte in the unspiked sample;
• Plots of values of blank determinations on a segmented
chart
• This is not true CC because this is no control limit on value;
• The chart is simply a plot oh the responses sequential obtain, with
zero as the base line value.
Interpretation of Control Chart
• Interpreting X-chart
• One or more points outside the three standard deviation limit (3s);
or
• Two or more consecutive points outside the two standard deviation
limit (2s); or
• A series of seven or more points above or below the mean; or
• An increasing or decreasing trend.
• Interpreting R-chart
• Similar to X-chart, five or more consecutive point above the 50%
confidence limit indicates a tendency toward the process being out
of control.
