WHY ?
 Tujuan

Untuk mengetahui perlakuan terbaik pemberian bahan anastesi ekstrak

hati batang pisang dalam sistem transportasi basah ikan Gurame.

Hipotesis

1. Ho : µ=0 : Pemberian ekstrak hati batang pisang tidak berpengaruh

terhadap anastesi ikan Gurame (Osphronemus gouramy)
pada sistem transportasi basah.
2. Ho : µ≠0 : Pemberian ekstrak hati batang pisang berpengaruh
terhadap anastesi ikan Gurame (Osphronemus gouramy)
pada sistem transportasi basah.

Waktu dan Tempat
20 Juli 2020-3 Agustus 2020 bertempat
di Laboratorium Akuakultur UNIHAZ
Bengkulu dan dengan rute perjalanan
kota Bengkulu-Kepahiyang-Curup-Lebon-
kota Bengkulu
Alat dan Bahan
Alat: Box, plastik 20L, thermometer, pH
meter, water teskit, pipa
volumetrik dan mobil.
Bahan: Ikan Gurame,Ekstrak hati batang
pisang dan pakan ikan.
Rancangan Percobaan
Rancangan Acak Lengkap (RAL)
Terdiri dari 3 perlakuan dan 5 ulangan
P1: Dosis dengan konsentrasi ekstrak 5% (v/v)
P2: Dosis dengan Konsentrasi ekstrak 10% (v/v)
P3: Dosis dengan Konsentrasi ekstrak 15% (v/v)

5 Respon dan tingkah laku ikan

Setiap perlakuan menunjukkan bahwa saat
dilakukan anastesi ikan melalui beberapa
tahapan dimulai dari fase normal hingga fase
pingsan.
Kualitas air
Setelah uji coba transportasi kualitas
air mengalami penuruan drastis.
Pembahasan

kelangsungan hidup ikan Gurame setiap perlakuannya

menghasilkan nilai yang berbeda. Setelah dilakukan
analisis sidik ragam didapatkan nilai F. Hitung lebih besar
dibanding dengan F. tabel 5% dan 1% ini berarti bahwa
pemberian ekstrak hati batang pisang sebagai bahan
anastesi berbeda berpengaruh sangat nyata terhadap
kelangsungan hidup ikan Gurame. Kemudian dilakukan uji
lanjut BNT pada taraf 5% dan 1% didapatkan P2 tidak Kelangsungan
berbeda dengan P3 tetapi berbeda dengan P1. hidup

Diantara P1, P2 dan P3 didapatkan nilai kelangsungan hidup ikan tertinggi dihasilkan pada P1
dengan konsentrasi 5% didapatkan kelulusan hidup rata-rata sebesar 78%. Kelulusan hidup ikan
Gurame terkecil didapatkan pada ekstrak hati batang pisang dengan konsentrasi 15 %. Hal ini
diduga bahwa saat ikan diberikan anestesi ikan menjadi shock karena perubahan lingkungan
sehingga ikan melakukan gerakan yang berlebihan. Pada proses shock teersebut menyebabkan ikan
mengalami kematian karena pada kondisi tersebut ikan yang stres akan terjadi peningkatan asam
laktat dalam darah (Pratisari 2010). Pada konsentrasi yang tinggi, kandungan bahan kimia di hati
batang pisang juga tinggi seperti saponin. Saponin juga bersifat bisa menghancurkan butir darah
merah lewat reaksi hemolisis, bersifat racun bagi hewan berdarah dingin, dan banyak diantaranya
digunakan sebagai racun ikan (Cheek 2005).
Pembahasan

Pengamatan masa induksi menunjukkan bahwa perlakuan dengan konsentrat

15% memiliki waktu tercepat dalam pemingsanan dengan rata-rata waktu
pingsan 66 menit. Hal ini diduga dikarenakan dosis ekstrak hati batang pisang
yang digunakan lebih banyak dari perlakuan yang lainnya. Sedangkan waktu
terlama dalam pemingsanan ikan Gurame terjadi pada perlakuan dengan
konsentrasi 5% yaitu dengan rata-rata waktu 102menit.

Pada pengamatan lama waktu sedatif dihitung pada saat ikan

berada di dalam wadah penyadaran yang diaerasi. Ciri-ciri ikan
telah sadar adalah ikan telah mulai berenang normal dan
menerima respon ransangan dari luar. Hasil pengamatan
menunjukkan bahwa waktu penyadaran ikan berbeda di tiap
perlakuan. Pada perlakuan 1 dengan konsentrasi 5%, ikan mulai
sadar di menit ke 7. Perlakuan 2 dengan konsentrasi 10%, ikan
mulai sadar di menit ke 14. Sedangkan waktu sadar terlama
berada di perlakuan dengan konsentrasi 15% yatiu ikan mulai
sadar di menit ke 20.
 Pembahasan

Berdasarkan tabel 6 pemberian ekstrak hati
batang pisang menunjukkan pengaruh terhadap
ikan yang diujikan. Pengaruh yang diberikan
tersebut dapat dilihat dari gerakan operkulum
yang mulai melemah, sirip punggung yang
meregang, sesekali mulut disembulkan ke
permukaan serta sebagian ikan memasuki fase
pingsan ringan dan pingsan berat. Perubahan
aktivitas ikan uji mulai terlihat pada menit ke-30
hingga menit ke-60. Pada perlakuan konsentrasi 5
% memasuki tahap pingsan pada menit ke 101,
102 dan 104. Perlakuan konsentrasi 10 %
memasuki tahap pingsan pada menit ke 73,74,
dan 75, sedangkan pada perlakuan konsentrasi 15
% ikan memasuki tahap pingsan pada menit ke
63, 65, 66, dan 68.
Saat dilakukan anastesi ikan melalui beberapa
tahapan dimulai dari fase normal hinggan fase
pingsan. Fase normal adalah fase dimana ikan
masih reaktif terhadap rangsangan luar,
pergerakan operculum dan kontraksi otot normal
selanjutnya ikan memasuki fase kehilangan
keseimbangan. Fase ini ikan mengalami kontraksi
otot lemah, berenang tidak teratur memberikan
reaksi hanya terhadap rangsangan getaran dan
sentuhan yang sangat kuat dan pergerakan
operculum cepat. Fase pingsan ringan ikan mulai
mengalami reaktifitas terhadap rangsangan luar
sedikit menurun, pergerakan operculum
melambat, keseimbangan normal (Tidwel et.al
2004). Ikan memasuki fase pingsan ringan saat
tidak mengalami reaktivitas terhadap rangsangan
luar, kecuali dengan tekanan kuat. Pergerakan
operculum lambat, keseimbangan normal

Pratisari (2010) menyatakan ikan yang mengalami fase pingsan ringan, pingsan berat dan roboh memiliki tingkat
respirasi dan metabolisme yang rendah. Dari saat ikan mengalami pingsan ringan sampai pingsan, pengaruh
konsentrasi pada perlakuan 10 % dan 15 % tidak menunjukan perbedaan yang nyata secara visual hal ini diduga dosis
yang diberikan sudah cukup untuk mempengaruhi sistem syaraf ikan. Pemberian dosis yang berlebih akan
menyebabkan kerusakan sistem syaraf dan akan berakibat overdosis atau kematian (Arliansah 2009).
 Kesimpulan

1. Hasil penelitian menunjukan bahwa penggunaan ekstrak hati batang pisang

sebagai bahan anastesi berpengaruh sangat nyata terhadap kelangsungan
hidup ikan Gurame (Osphronemous gourame) pada sistem transportasi
basah tertutup.
2. Berdasarkan hasil uji lanjut BNT didapatkan Perlakuan dosis konsentrasi 5%
(P1). Merupakan perlakuan terbaik dibandingkan dengan perlakuan lainnya.
 Saran

Dari penelitian yang telah dilakukan penulis menyarankan

penggunaan bahan anastesi ekstrak hati batang pisang dengan
konsentrasi 5% untuk transportasi basah tertutup ikan Gourame.
