 PENGGOLONGAN
Senyawa tanin termasuk ke dalam senyawa polifenol yang
artinya senyawa yang memiliki bagian berupa fenolik.
Senyawa tanin dibagi menjadi dua berdasarkan pada sifat
dan struktur kimianya, yaitu tanin yang terhidrolisis dan
tanin yang terkondensasi. Tanin terhidrolisis biasanya
ditemukan dalam konsentrasi yang lebih rendah pada
tanaman bila dibandingkan dengan tanin terkondensasi.
Tanin terkondensasi terdiri dari beberapa unit flavanoid
(flavan-3-ol) dihubungkan oleh ikatan-ikatan karbon. Tanin
terkondensasi banyak ditemukan dalam berbagai jenis
tanaman seperti Acacia spp, sericea Lespedeza serta spesies
padang rumput seperti Lotus spp.

biasanya tidak dapat dihidrolisis, tetapi dapat
terkondensasi menghasilkan asam klorida.

Tanin jenis ini kebanyakan terdiri dari
polimer flavonoid yang merupakan senyawa
fenol. Nama lain dari tanin ini adalah
Proanthocyanidin. Proanthocyanidin
merupakan polimer dari flavonoid yang
dihubungkan dengan melalui ikatan C-8
dengan C-4. Salah satu contohnya adalah
Sorghum procyanidin
Tanin terhidrolisis biasanya berikatan
dengan karbohidrat dengan
membentuk  jembatan oksigen, maka
dari itu tanin ini dapat dihidrolisis
dengan menggunakan asam sulfat atau
asam klorida. Salah satu contoh jenis
tanin ini adalah gallotanin yang
merupakan senyawa gabungan dari
karbohidrat dengan asam galat.
 STRUKTUR KIMIA
Tanin atau lebih dikenal dengan asam tanat, biasanya mengandung 10% H2O. Struktur kimia tanin adalah kompleks dan
tidak sama. Asam tanat tersusun 5 - 10 residu ester galat, sehingga galotanin sebagai salah satu senyawa turunan tanin
dikenal dengan nama asam tanat. Beberapa struktur kimia senyawa tanin adalah sebagai berikut.

Tanin Terkondensasi, Proanthocyanidin merupakan polimer

dari flavonoid yang dihubungkan dengan melalui ikatan C-
8 dengan C-4.
 Rantai ester poligallol ditemukan di dalam gallotanin
terbentuk dari meta-atau para-depside obligasi, melibatkan
hidroksil fenolik daripada gugus hidroksi alifatik. Depside
obligasi lebih mudah dihidrolisis daripada ikatan ester
alifatik. Metanolisis dalam asam lemah dengan
menggunakan metanol dapat menghancurkan depside
tetapi tidak ester obligasi. Dengan demikian poliol inti
dengan kelompok galloyl yang teresterisasi dapat
dihasilkan dari campuran kompleks dari ester polygalloyl
oleh metanolisis dengan buffer asetat. Asam kuat mineral,
panas, dan metanol dapat digunakan untuk metanolisis
baik depside dan ester obligasi menghasilkan poliol inti
dan metil galat. Hidrolisis dengan asam kuat dapat
mengubah galotanin menjadi asam galat dan poliol inti.
TANAMAN PENGHASIL
Jenis tanaman yang mengandung tanin antara lain
adalah daun sidaguri (Sida rhombifolia L.) yang
diketahui mengandung tanin cukup tinggi dan telah
digunakan sebagai pestisida nabati pembunuh ulat
(larvasidal) (Kusuma et al., 2009; Islam et al., 2003).
Daun melinjo (Gnetum gnemon L.) juga mengandung
tanin. Daun gamal (Gliricidia sepium Jacq.) dan lamtoro
(Leucaena leucocephala Lamk.) mempunyai kandungan
tanin 8-10% (Suharti, 2005; Sulastri, 2009). Biji pinang
(Areca catechu L.) dan simplisia gambir (Uncaria
gambir Roxb.) telah dikenal luas sebagai penghasil
tanin dengan kandungan tanin masing-masing sebesar
26,6% dan 30-40%
 Tanaman sirih merah
(Piper crocatum)
termasuk dalam famili Piperaceae, tumbuh
merambat dengan bentuk daun menyerupai
hati dan bertangkai, yang tumbuh
berselang-seling dari batangnya serta
penampakan daun yang berwarna merah
keperakan dan mengkilap. Dalam daun
sirih merah terkandung senyawa fitokimia
yakni alkoloid, saponin, tanin dan
flavonoid.
Daun dewa (Gynura
divaricata)
mengandung zat saponin, minyak atsiri,
flavonoid, dan tanin. Efek farmakologis daun
dewa adalah antikoagulan (koagulan=zat yang
mempermudah dan mempercepat pembekuan
darah), mencairkan bekuan darah,
stimulasi sirkulasi, menghentikan perdarahan,
menghilangkan panas, dan membersihkan
racun.
Buah, daun, dan kulit batang pohon
jambu biji (Psidium guajava)
mengandung tanin, sedang pada
bunganya tidak banyak
mengandung tanin. Daun jambu
biji juga mengandung zat lain
kecuali tanin, seperti minyak
atsiri, asam ursolat, asam
psidiolat, asam kratogolat, asam
oleanolat, asam guajaverin dan
vitami

Tumbuhan ini mempunyai kandungan kimia
berupa chlorogenik acid, asam citrun, fisalin,
flavonoid, saponin, polifenol. Buah
mengandung asam malat, alkaloid, tanin,
kriptoxantin, vitamin C dan gula.Biji
mengandung elaidic acid. Sifat tumbuhan ini
analgetik (penghilang rasa sakit), peluruh air
seni (diuretik).
