Sejarah penelitian enzim tanah
• Louis Pasteur tahun 1860 menyatakan bahwa fermentasi dikatalisir
oleh enzim yang berkaitan erat dengan struktur dan proses hidup
dari sel ragi
• Edward Buchner tahun1897 memperlihatkan bahwa enzim terlibat
dalam fermentasi alkohol yang dapat diekstrak dari sel ragi dan
dapat berfungsi secara terpisah dari sel
• Pertama kali dilaporkan oleh WOODS (1899) pada Annual Meeting
of The American Association for the Advancement of Science in
Columbus OHIO.
• Namun demikian sedikit sekali kemajuan dicapai sampai tahun 1950
• Berbagai reaksi enzimatik diperoleh baru setelah tahun 1950
• Sejarah tentang sejarah penelitian enzim tanah ditulis oleh
SKUJINS (1978).

3. PERAN ENZIM TANAH DALAM
PEMBENTUKAN BAHAN HUMIK
• Proses pembentukan substansi humik merupakan salah satu hal yang
sangat sedikit diketahui dalam aspek kimia humus dan salah satu hal yang
penuh perdebatan. Penelitian tentang hal ini terus berlanjut dalam usaha
mengungkap proses tersebut berdasarkan teori dan temuan yang ada di
lapangan.
• Beberapa lintasan (pathway) dalam pembentukan bahan humik selama
perombakan (dekomposisi) sisa-sisa tanaman dan hewan dalam tanah
telah diperkenalkan. Beberapa teori yang terkenal adalah :
Teori klasik yang dipopulerkan Waksman, yang menyatakan bahwa bahan
humik merupakan lignin yang termodifikasi (pathway 1)
namun demikian mayoritas peneliti dewasa ini lebih cendrung menerima
mekanisme yang melibatkan quinones (pathway 2 and 3). Dalam praktek
pathway ke-empatmerupakan proses atau mekanisme yang dapat dianggap
menyerupai proses sebenarnya dalam sintesis asam humat dan asam
fulvat di alam termasuk kondensasi gula-amine (pathway 4).
• Keempat proses atau mekanisme ini mungkin terjadi di semua tanah
tergantung kepada faktor yang mempengaruhi. Pathway lignin mungkin
dominan pada tanah yang jelek drainasenya dan tanah alluvial (gambut,
rawa) sementara pathway polyphenols lebih utama terjadi pada tanah
hutan. Flukstuasi temperatur, kelembaban dan penyinaran yang tajam dan
sering seperti pada iklim kontinental mungkin cendrung mengikuti
mekanisme kondensasi gula-amine.

• The following evidence was cited by Waksman in support of the
lignin theory of humic acid formation:
• Both lignin and humic acid are decomposed with considerable
difficulty by the great majority of fungi and bacteria.
• Both lignin and humic acid are partly soluble in alcohol and pyridine.
• Both lignin and humic acid are soluble in alkali and precipitated by
acids.
• Both lignin and humic acid contain OCH3 groups.
• Both lignin and humic acid are acidic in nature.
• When lignins are warmed with aqueous alkali, they are transformed
into methoxyl-containing humic acids.
• Humic acids have properties similar to oxidized lignins.

mikroorganisme dari bahan non lignin sebagai
sumber C (e.g., celulose). Polyphenols
dioksidasi secara enzimatik menjadi quinones

humik.
• Dewasa ini teori klasik sudah ditinggalkan oleh
peneliti. Konsep yang menyatakan quinone dan
lignin keduanya disintesis oleh mikroorganisme
dan merupakan unit dasar dari bahan humik
lebih bisa diterima.

• Pembentukan substansi coklat sebagai reaksi
yang melibatkan quinones bukanlah peristiwa
yang langka sebaliknya dikenal luas dalam
pembentukan melanine, seperti halnya pada
daging buah yang masak dan sayuran sebagai
lanjutan dari kerusakan fisik dan selama proses
pembentukan lapisan pelindung biji.
• Kemungkinan sumber phenol untuk
pembentukan humus adalah dari lignin,
microorganisme, phenol dalam tanaman tannin.
Dalam hal ini kemungkinan terbesar adalah dari
lignin.

4. Karakteristik bahan humik
• Asam humat- fraksi humik yang tidak larut dalam air
pada kondisi masam (pH < 2) tetapi larut pada pH tinggi
(alkalis). Dapat diekstrak dari tanah dengan
menggunakan berbagai pelarut tetapi tidak larut dalam
asam. Asam humat merupakan komponen terbanyak
dari bahan humik yang dijumpai. Berwarna coklat gelap
sampai hitam.
• Asam fulvat- fraksi humik yang larut dalam air pada
kondisi pH masam amupun alkalis. Tetap berada dalam
larutan setelah pengendapan asam humat melalui
pengasaman larutan. Asam Fulvat berwarna kuning
sampai kuning kecoklatan.
• Humin – fraksi humik yang tidak larut dalam air baik
pada pH masam ataupun alkalis. Humin berwarna hitam.

4. Karakteristik bahan humik
• The low - molecular - weight fulvic acids have higher oxygen but
lower carbon contents than the high - molecular - weight humic
acids. Fulvic acids contain more functional groups of an acidic
nature, particulary COOH. The total acidities of fulvic acids (900 -
1400 meq/100g) are considerably higher than for humic acids (400 -
870 meq/100g).
