Stabilitas obat => berbagai factor yang dapat mempengaruhi stabilitas sediaan obat dan bagaimana cara
meningkatkan kestabilannya selama penyimpanan dan masa edarnya.
1. Faktor Lingkungan
- Suhu
- Oksigen: terjadi reaksi oksidasi. Diberikan antioksidan untuk mencegah
- Karbondioksida
- Cahaya
- Kelembaban
Degradasi obat melalui rute hidrolisis: biasanya untuk obat yang dilarutkan dalam air atau
mengandung air sehingga zat zat tersebut dapat mengalami reaksi penguraian jika terkena air
Degradasi melalui rute oksidasi: dengan senyawa lain atau dengan udara
Degradasi obat melalui fotolisis: untuk senyawa yang tidak stabil terhadap cahaya, harus ditaruh
tempat tidak terkena cahaya.
Reaksi kompleks degradasi obat: bisa terjadi dengan berbagai senyawa lain yang terkandung
dalam suatu formulasi

KINETIK DAN ORDE REAKSI
Ada 2 pendekatan:
o TERMODINAMIKA = membahas apa yang menyebabkan suatu reaksi itu terjadi dan
apa yang menyebabkan kestabilan itu berubah, berhubungan dengan energi,
o KINETIKA = berhubungan dengan proses perubahan nya energy, sehingga terjadinya
reaksi, apakah reaksi nya cepat atau lambat
Kinetika berhubungan dengan pergerakan molekulnya (kimia)
Yang mempengaruhi derajat perubahan nya, (mana yang laju nya cepat mana yang lambat =
orde)
o Stabilitas
o Inkompatibilitas

o Kerja obat pada level molecular

Kinetika berkaitan dengan studi tentang laju terjadinya proses dan mekanisme reaksi kimia

2. Half-life method
● This method is based on the relationship between the initial concentration of the reactant, the
half life, and the reaction order.
● For zero-order reactions, t1/2 increases with increasing concentration, whereas for first-order
reactions, t1/2 does not change with change in concentration

of the reacting species over a wide range of reactions. (gajelas karna dia dipertengahan jadi perlu

● These may occur under the following conditions:
➢ One or more of the reactants enters into the rate equation in great excess compared to others;

➢ One of the reactant is catalyst; adanya katalis
➢ One or more of the reactants is constantly replenished during the course of reaction adanya suatu

If one dose of aspirin at 650 mg per teaspoonful is administered, what is the shelflife of suspension?
Increasing the temperature increases reaction rates because of the disproportionately large
increase in the number of high energy collisions (peningkatan yang tidak proporsional dalam
jumlah tabrakan energi yang tinggi). It is only these collisions (possessing at least the activation
energy for the reaction) which result in a reaction.
